For breeding of paphiopedilum, 15 varieties and species were collected and 5 were renewed.1 hybrid was selected as a potential hybrid with bright yellow green flower color, and flowering period was August-September. In this study, we also tried to improve The tissue culture methods of immature flower bud of Paphiopedilum, the average survival rate of the test three varieties was 11.5-53.1%. The optimal subculture conditions were light / dark at 10h / 14h, and supplement with 18.8 and 37.6 mM potassium nitrate (KNO 3 ) as nitrogen source in medium. In addition, using shoot tip as explants to cultured in medium supplement with kinetn, there are 7.5-30.0% of explants formed callus.As for developing flower regulation system of paphiopedilum, GA would cause serious malformation no matter paclobutrazol (PBZ) added when or not.While added GA for inducing flowering of complex 4266, it was showed that GA could accelerate flowering but also caused malformation of flowers.Considering flower induction of nobile type dendrobium, ‘the total flower buds number’, ‘number of nodes with flower buds’ of Lai's Sunnyboy and Lai's Lovely Pearl could be increased when induced flowering with ‘Technique for Induction Flowering of Dendrobium Nobile-type’which developed byTSIPS accompanied different growth retardants. The best treatment for Lai's Sunnyboy was 25℃/25℃ combined with CCC 100ppm while 25℃/15℃ combined with PBZ 5ppm was the best for Lai's Lovely Pearl.
