Study of Inspection and Monitoring System for Gene-edited and Genetically Modified crops

The main goal of this project is to enhance Taiwan's capacity for monitoring and testing agricultural biotechnology crops. A qualitative and quantitative detection method for genetically modified soybean (DBN-09004-6) has been developed, with a detection limit of 0.05%. In optimizing the GM potato detection system, the "CTAB extraction" method performed best, while the tested "automated extraction-reagent 2" shows significant improved efficiency of nucleic acid extraction, offering a viable alternative for automated systems currently used. In proficiency testing, most laboratories achieved 100% accuracy for blind samples of papaya, maize, and soybean, with only Lab B reporting false negative results (NPTII, PRSV-CP) for papaya. In the external proficiency test held by FAPAS, our analysis results are all consistent with the standard answers. Furthermore, in the 2024 testing and monitoring activities for genetically modified crops, 36 samples retrieved from investigated fields (31 potatoes, 4 maize, 1 soybean) and 100 samples collected from imported crops (56 soybean, 43 maize, 1 cotton) are analyzed.

Note: In the figure above, "M" represents the marker, and "ddH2O" refers to double-distilled water. In Figure 1 (left), PCR amplification products of genetically modified soybean event (DBN-09004-6) at various concentrations (100%, 1%, 0.1%, 0.05%) are shown (target gene: Le1; 74 bp). In Figure 1 (right), PCR amplification products of genetically modified soybean event (DBN-09004-6) at different concentrations (100%, 1%, 0.1%, 0.05%) are shown (target gene: DBN-09004-6 detection fragment; 89 bp).
